ABSTRACT

Objective To investigate the adults smoking status of Hui and Han in Ningxia. Methods Among 6 monitoring sites in Ningxia from 2013 to 2014, multi-stage stratified cluster random sampling method was used, information about risk factors of chronic diseases by were collected questionnaire investigation, a total of 3 527 subjects were effective sample for smoking analysis among 3 540 adults aged 18 and over. After the complex weighting, the smoking status of different nationalities, sexes and age was analyzed. Results The current smoking rate was 29.42% (95% CI:27.91%-30.92%), the current smoking rate of male(56.73%, 95% CI:54.25%-59.20%) was higher than female (0.92%, 95% CI:0.50%-1.34%) (P<0.001); The current smoking rate of Han (32.04%, 95% CI:30.24%-33.85%) was higher than Hui (23.09%, 95% CI:20.41%-25.76%). The daily smoking rate of Han (27.98%, 95% CI: 26.25%-29.72%) was higher than that of Hui nationality (19.83%, 95% CI:17.30%-22.36%) (P=0.001), and that of male (49.41%, 95% CI: 46.92%-51.91%) was higher than that of female (0.73%, 95% CI: 0.36%-1.11%) (P<0.001). The average daily smoking of male (20) was higher than that of female (8) (Z=-4.448, P<0.001). Smokers quit smoking rate was 12.54% (95% CI:10.50%-14.57%). Adult secondhand smoke exposure rate was 54.44% (95% CI:52.53%-56.36%). Conclusions The smoking rate of adult residents is high in Ningxia province, but the quit smoking rate is low, male smoking rate is higher than female, Han smoking rates higher than the Hui. The sample population was highly exposed to secondhand smoke. Tobacco control interventions should be taken against high-risk groups.

ABSTRACT

In the present study, we examined the effect of oxygen glucose deprivation (OGD) post-conditioning (PostC) on neural cell apoptosis in OGD-PostC model and the protective effect on primary cortical neurons against OGD injury in vitro. Four-h OGD was induced by OGD by using a specialized and humidified chamber. To initiate OGD, culture medium was replaced with de-oxygenated and glucose-free extracellular solution-Locke's medium. After OGD treatment for 4 h, cells were then allowed to recover for 6 h or 20 h. Then lactate dehydrogenase (LDH) release assay, Western blotting and flow cytometry were used to detect cell death, protein levels and apoptotic cells, respectively. For the PostC treatment, three cycles of 15-min OGD, followed by 15 min normal cultivation, were applied immediately after injurious 4-h OGD. Cells were then allowed to recover for 6 h or 20 h, and cell death was assessed by LDH release assay. Apoptotic cells were flow cytometrically evaluated after 4-h OGD, followed by re-oxygenation for 20 h (O4/R20). In addition, Western blotting was used to examine the expression of heat-shock protein 70 (HSP70), Bcl-2 and Bax. The ratio of Bcl-2 expression was (0.44±0.08)% and (0.76±0.10)%, and that of Bax expression was (0.51±0.05)% and (0.39±0.04)%, and that of HSP70 was (0.42±0.031)% and (0.72±0.045)% respectively in OGD group and PostC group. After O4/R6, the rate of neuron death in PostC group and OGD groups was (28.96±3.03)% and (37.02±4.47)%, respectively. Therefore, the PostC treatment could up-regulate the expression of HSP70 and Bcl-2, but down-regulate Bax expression. As compared with OGD group, OGD-induced neuron death and apoptosis were significantly decreased in PostC group (P<0.05). These findings suggest that PostC inhibited OGD-induced neuron death. This neuro-protective effect is likely achieved by anti-apoptotic mechanisms and is associated with over-expression of HSP70.

ABSTRACT

<p><b>OBJECTIVE</b>To observe the osteoinductive activity of demineralized bone matrix (DBM) and deproteinized bone (DPB) made from human avascular necrotic femoral head.</p><p><b>METHODS</b>The femoral head was cut into pieces with the size of 3 mm x 3 mm x 5 mm, which were made into DBM and DPB. These two kinds of biomaterials were cocultured with human bone mesenchymal stem cells (hBMSCs). Monolayer cells without biomaterials were cultured as control. Proliferative activity of hBMSCs was evaluated on days 1, 3, 5, 7 and 14. The concentration of alkaline phosphatase (ALP), osteocalcin (OC), and Ca(2+) were detected on days 1, 7, 14 and 21.</p><p><b>RESULTS</b>Cells cultured in DBM showed higher proliferative activity than did in DPB and monolayer cells (F =39.773, P <0.01). DBM and DPB also had osteoinductive activity. The concentrations of ALP (F=93.162, P <0.01), OC (F =236.852, P < 0.01), Ca(2+)(F =80.711, P <0.01) of DBM group were significantly higher than that of DPB and control groups.</p><p><b>CONCLUSIONS</b>In vitro, DBM and DPB made from avascular necrotic femoral head have osteoinductive activity when cocultured with hBMSCs, and the former is stronger than the latter.</p>

ABSTRACT

Objective To know the situation of tobacco advertisement, promotions and related factors in six cities in China. Methods 4815 adults (above 18 years), selected form Beijing, Shanghai,Shenyang, Changsha, Guangzhou and Yinchuan through probability proportionate sampling and simple random sampling, were investigated through questionnaires. Results The most commonly reported channels that smokers noticed tobacco advertisements were billboards (35.6%) and television (34.4%).The most commonly reported tobacco promotional activities that were noticed by smokers were flee gills when buying cigarettes (23.1% ) and free samples of cigarettes (13.9%). Smokers in Changsha were more likely to report noticing tobacco advertisement on billboards (X2=562.474, P<0.001), and on television(X2=265.570, P<0.001 ). Smokers in Claangsha (X2=58.314, P<0.001 ) were more likely to notice tobacco related news and games. A logistic regression analysis showed that the living and education level were related to awareness of tobacco advertisement and promotion. Conclusion It was universal to see tobacco advertisement and promotions in cities in China but the laws and regulations about tobacco-control were not uniformly executed in different cities. It is necessary to perfect and uniform related laws and regulations.

ABSTRACT

<p><b>OBJECTIVE</b>To establish a method for SNP genotyping of multi-genes by allele-specific oligonucleotide probe ligation mediated by a thermostable ligase, and to explore the genetic polymorphisms of drug-metabolizing enzymes in breast cancer patients and their association with chemotherapeutic responses.</p><p><b>METHODS</b>10 SNP loci of enzyme genes related to chemotherapeutic drugs such as taxanes, anthracyclines and cyclophosphamide were selected, and were genotyped for blood samples from 126 breast cancer patients by the established method. Their correlations with therapeutic responses were retrospectively evaluated.</p><p><b>RESULTS</b>The lower detection limit of genomic DNA by this developed method was 6.25 ng. The fluorescent peak locations of ligation products on ABI PRISM 377 DNA sequencer were accurate and consistent with prospective sizes in bases (Bias range 0.08 - 0.69 bp, x(-) = 0.31 bp, s = 0.18 bp). Same genotyping results were obtained for repeat tests of 8 random samples, which were further confirmed by sequencing analysis. The 10 SNP loci were polymorphic of different frequency in the breast cancer patients. The combinations with GSTP1 genotypes and GSTM1 genotypes were related to anthracycline-based chemotherapy efficacy (P = 0.037), and the low GSTs activity group (GSTP1 variant allele + GSTM1 null) showed the best effects (85.7%). GSTM1 genotypes and their combinations with GSTP1 and/or CYP3A5*3 genotypes were related to taxane-based therapy efficacy (P < 0.05 for all), and both the low GSTs activity group and the drug slow-metabolising group (low GSTs activity group + CYP3A5*3 wild allele) showed better effects (100%).</p><p><b>CONCLUSION</b>The established method is reliable and applicable in multiplex SNPs genotyping of multi-genes. SNPs combination may have a better clinical application value for prediction of chemotherapeutic responses.</p>

ABSTRACT

<p><b>AIM</b>To investigate the expression of cyclooxygenase-2 (COX-2) and epidermal growth factor receptor (EGFR) and the possible mechanism in the development in androgen independent prostate cancer (AIPC).</p><p><b>METHODS</b>Immunohistochemistry was performed on paraffin-embedded sections with goat polyclonal against COX-2 and mouse monoclonal antibody against EGFR in 30 AIPC and 18 androgen dependent prostate cancer (ADPC) specimens. The effect of epidermal growth factor (EGF) treatments on the expression of COX-2 and signal pathway in PC-3 and DU-145 cells was studied using reverse transcription-polymerase chain reaction (RT-PCR) and Western blot analysis. ELISA was used to measure prostaglandin E2 (PGE2) levels in the media of PC-3 and DU-145 incubated with EGF for 24 h.</p><p><b>RESULTS</b>COX-2 was positively expressed in AIPC and ADPC, which were predominantly in endochylema of prostate cancer (PCa) cells. Intense staining was seen in AIPC (80%) and in ADPC (55.5%), but there was no significant association between the two groups. EGFR expression was also positive in the two groups (61.8% in ADPC and 90% in AIPC, P < 0.01). A significant association was found between EGFR expression and a higher Gleason score (P < 0.05) or tumor stage (P < 0.05). The expression of PGE2 was increased in PC-3 and DU-145 cells after being incubated with EGF. Both p38MAPK and PI-3K pathway were involved in the PC-3 cell COX-2 upregulation course. In DU-145, only p38MAPK pathway was associated with COX-2 upregulation.</p><p><b>CONCLUSION</b>EGFR activation induces COX-2 expression through PI-3K and/or p38MAPK pathways. COX-2 and EGFR inhibitors might have a cooperative anti-tumor effect in PCa.</p>

ABSTRACT

<p><b>BACKGROUND</b>This study aimed to investigate multi-slice CT contrast-enhanced presentation of gastric cancer and its correlation with histo-differentiation and p53 and P-glycoprotein (P-gp) expression.</p><p><b>METHODS</b>Sixty-six patients with gastric cancer in the present study underwent a multi-slice CT preoperative routine and dual-phase contrast-enhanced examination of the upper abdomen; postoperative specimens were used to determine histo-differentiation and the expression of p53 and P-gp. The correlation of multi-slice CT contrast-enhanced presentation with histo-differentiation and expression of p53 and P-gp was analyzed.</p><p><b>RESULTS</b>The dual-phase contrast-enhanced ratio (CER) was not correlated with the histo-differentiation of gastric cancer (P > 0.05). Positive expression of p53 and P-gp was significantly higher in the cases of layered or heterogeneous enhancement than in the cases of homogenous enhancement (P < 0.05). Positive expression of p53 was also correlated with the arterial phase CER, tumor size and lymph node metastasis (P < 0.05), but not with infiltration thickness of the gastric wall, nor was it correlated with the portal phase CER (P > 0.05). Positive expression of P-gp was only correlated with the portal phase CER (P = 0.005).</p><p><b>CONCLUSIONS</b>Differently enhanced pattern and CER of the arterial and portal phase in gastric cancer correlate with its different histo-differentiation and expression of p53 and P-gp respectively. In addition, tumor size and lymph node metastasis of gastric cancer relate to the expression of p53.</p>

ABSTRACT

<p><b>OBJECTIVE</b>To study the changes in circulating VEGF and endostatin (ES) levels during chemotherapy for patients with breast cancer, and their correlation with efficacy of chemotherapy.</p><p><b>METHODS</b>40 breast cancer patients with metastases were included in this study. They received TAC/TEC, CAF/CEF, NP, CAP, CMF, TFP, TA or TC regime chemotherapy, respectively. Totally 120 serum samples were collected from the patients at three time points: before chemotherapy, the end of 1 and 5-6 chemotherapy cycles, and analyzed for VEGF and ES levels using ELISA. Tumor agiogenesis activity was evaluated by serum soluble vascular cell adhesion molecule (VCAM - 1) measured by ELISA as a surrogate marker.</p><p><b>RESULTS</b>(1) Before chemotherapy, the median level of VEGF in patients with breast cancer was 496.6 pg/ml, 4.7 times higher than that of healthy controls (P <0.001). The median level of ES was 95.5 ng/ml, 18.3% lower than that of healthy controls (P = 0.183). VCAM-1 was 1077.1 ng/ml and higher than that of controls (P <0.001). The serum VEGF levels correlated with VCAM-1 levels, tumor staging and metastatic sites (P <0.05). (2) At the end of 1 cycle of chemotherapy, the serum VEGF level (median 524.8 pg/ml) was higher than the pretreatment values (P = 0.047), whereas the levels of ES and VCAM-1 were not significantly altered (110.5 ng/ml, P = 0.055; and 975.6 ng/ml, P = 0.27). (3) At the end of 5-6 cycles, the changes in VEGF correlated with the response to chemotherapy. Serum VEGF levels in 27 patients with chemotherapy-responsive and stable disease showed a significant decrease (median 287.4 pg/ml) , but not observed in 13 patients with progressive disease. VCAM-1 also showed a treatment-related change like VEGF. However, chemotherapy might only have a minor effect on ES, because there was no significant difference in the ES levels among 5-6 cycle patients, 1 cycle patients and healthy controls, and neither between therapy-responsive patients.</p><p><b>CONCLUSION</b>Intensive chemotherapy for breast cancer results in a significant decrease of serum VEGF level, which might be an indicator of the controlled disease status, and following the treatment-induced response or stabilization, the tumor angiogenesis seems to change into an anti-angiogenesis direction.</p>

@#ObjectiveTo observe effect of head and body acupuncture and moxibustion on stroke.Methods183 stroke patients were randomly divided into 3 groups, head acupuncture and moxibustion, body acupuncture and moxibustion, and head body acupuncture and moxibustion. After two months treatment, effects of 3 groups were evaluated.ResultsThere were no differences between head group and body group, body group and head body group, but there was significantly difference between head body group and head group (P<0.05).ConclusionThe head body acupuncture and moxibustion can gain the best clinical effect on stroke patients compared with simply head or body acupuncture and moxibustion.

Objective To establish a desirable quantitative assay system to evaluate the correlation between surgical manipulation and micrometastasis in primary esophageal cancer patients.Methods 118 pe- ripheral blood samples from 40 esophageal cancer patients undergoing radical resection were collected before surgery(B-1),immediately after surgery(B0)and at the third day postoperatively(B+3).Based on real-time quantitative reverse transcription-PCR,carcinoembryonic antigen(CEA)mRNA gene was used in the detec- tion.12 patients with benign tumor and 13 healthy volunteers were collected as negative control groups.Re- sults The median of CEA mRNA copies at B-1,B0 and B+3 were 1592,13 314 and 6221 copies/ml blood, respectively.CEA mRNA expression levels were found to be significantly higher at both B0 and B+3 than that of B-1(P=0.0001 and 0.0209,respectively).No significant difference was found between B0 and B+3(P= 0.4396).Conclusion Surgical manipulation on esophageal cancer patients increases the probability of mi- crometastasis.Therefore,adjuvant therapy is needed during perioperative stage.
